Calf thymus polymerase.
نویسنده
چکیده
The term polymerase has been applied to the deoxyribonucleoside triphosphate-polymerizing enzyme isolated from Escherichia coli (1). Enzymes with similar activity have been purified from regenerating rat liver (2) and calf thymus gland (3). An ample and varied supply of such polymerases is desirable for studies on the mechanism of deoxyribonucleic acid synthesis. The present work describes a partial purification of the calf thymus polymerase and demonstrates the gross properties of the synthetic reaction. Certain useful properties of this enzyme preparation are currently being exploited for studies on the nature of primer DNA (4).

Purification of RNA polymerase IIO from calf thymus.
Three subspecies of RNA polymerase II, designated IIO, IIA, and IIB, have been described in calf thymus and shown to differ in the apparent molecular weight of their largest subunits, designated IIo, IIa, and IIb, respectively. The objective of this study was to develop a procedure for the purification of RNA polymerase IIO. This form of the enzyme predominates in vivo and is responsible for th...
متن کاملThe yeast analog of mammalian cyclin/proliferating-cell nuclear antigen interacts with mammalian DNA polymerase delta.
DNA polymerase III from Saccharomyces cerevisiae is analogous to the mammalian DNA polymerase delta by several criteria, including an increased synthetic activity on poly(dA).oligo(dT) (40:1 nucleotide ratio) in the presence of calf thymus proliferating-cell nuclear antigen (PCNA), or cyclin. This stimulation assay has been used to purify the yeast analog of PCNA/cyclin (yPCNA) to homogeneity. ...
متن کاملThe influence of syringomycin on ribonucleic acid synthesis.
Syringomycin, a wide-spectrum antibiotic produced by strains of Pseudomonas syringae which cause bacterial canker of peach, was able to bind to salmon sperm and calf thymus deoxyribonucleic acid but not to calf thymus histone; it also inhibited ribonucleic acid polymerase activity. These abilities to bind to deoxyribonucleic acid and to inhibit ribonucleic acid polymerase were inactivated when ...
متن کاملCalf thymus Hsc70 protein protects and reactivates prokaryotic and eukaryotic enzymes.
The heat-shock 70 protein (Hsp70) chaperone family is very conserved and its prokaryotic homologue, the DnaK protein, is assumed to form one of the cellular systems for the prevention and restoration of heat-induced protein denaturation. By using anti-DnaK antibodies we purified the DnaK homologue heat-shock cognate protein (Hsc70) from calf thymus to apparent homogeneity. This protein was clas...
متن کاملSingle-stranded DNA structure and DNA polymerase activity in the presence of nucleic acid helix-unwinding proteins from calf thymus.
In the preceding articles we have described the isolation and some of the properties of two calf thymus proteins which bind selectively to single-stranded DNA and which appear analogous to previously isolated prokaryotic DNA-unwinding proteins. In the present work we demonstrate two further points of analogy. First, both the calf UP1 and the high salt eluting proteins form protein-rich complexe...
